Dr. Wang’s research is focused on the interaction of host factors with RNA viruses. The first project of his group was to develop yeast as a model host, so that using reverse genetic approaches the replication of enterovirus 71 (EV71), and Japanese encephalitis virus could be studied in this simple eukaryotic organism. In another project, they will use proteomic approaches to analyze infection with EV71 in two cell lineages, RD cells and SF268 cells, focusing on the secreted proteins. First, by using one dimension electrophoresis and liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS), they will identify proteins which are secreted only by infected cells. Second, they will perform 2D-electrophoresis followed by MALDI-TOF in order to confirm the identity of those proteins. Once they identify the host proteins which are secreted by EV71 infected cells, they will evaluate the results obtained taking into account the proteins previously described in the secretome of dengue     virus-infected HepG2 cells, the proteins present in human plasma and proteins of interest for EV71 pathogenesis. Taken together the data will provide information critical for the understanding of the role of host protein secretion in disease progression.
